"CoVHIR-Vac: The VHIR vaccine design model for SARS-CoV-2"

Rapid diagnostics, vaccines and therapeutics are needed interventions to control the 2019 novel coronavirus (2019-nCoV) pandemic. Several vaccine candidates are in the pipeline. The likely earliest Phase 1 vaccine trial is a synthetic DNA-based candidate. Our Research group has a long record profile in preventive HIV vaccine development. Initially we constructed a chimerical Virus-like particles (VLPs) for Human papilloma virus and HIV vaccines. We have been using the yeast, baculovirus and eukaryotic expression system for VLPs production. Currently, we are focused in the SARS-CoV-2 vaccine development by using chimerical HPV/SARS-CoV-2 VLPs. We are using the L1 structural protein as a backbone and the S2 subunit of the spike glycoprotein as a target. For VLPs production we will use the eukaryotic expression system to elicite broad neutralizing antibodies. Thanks to the collaboration with Dr. Josep Quer, and the Hepatology laboratory research Group at VHIR, who sequenced two strains of SARS-CoV-2 from 2 patients isolates at Hospital Vall d’Hebron, we’ve confirmed that there is no consensus sequence variation between the strains isolated in our hospital and the Wuhan strains. This whole sequencing information will be useful to reach conserved regions as potential immunogens and targets for vaccine development and for eliciting broad neutralizing antibodies against pan-coronavirus. With the implementation of this project we will join the global effort to fight against this SARS-CoV-2 pandemic.
